Interference of Antioxidant Flavonoids with MTT Tetrazolium Assay in a Cell Free System

Introduction: The tetrazolium salt 3-(4, 5-dimethylthiazol-2-yl)-2, 5-diphenyltetrazolium bromide (MTT) used extensively to measure the quantitative survival and proliferation of mammalian cells. analysis is based upon reduction MTT by metabolically active cells insoluble formazan crystals. Flavonoids are a large group natural compounds found in plants with variable phenolic structures. Flavonoids, as they potential reducing agents, act free radical scavenger. aim study assess effect some flavonoids on their interference colorimetric MTT. cell viability obtained from assay was compared that SRB determination cytotoxicity. 
 Materials Methods: present examined few bio-flavonoids like Quercetin, EGCG, Rutin Resveratrol reduce absence under different experimental conditions such concentration flavonoids, incubation time results were findings. also involves flavonoid cytotoxicity lung cancer NCIH-460 NCIH-522 establish suitable for flavonoids.
 Results: All showed instant formation dark blue assay. Whereas cells, absorbance similar blank, indicating did not interfere cell-free system.
 Conclusion: From results, it evident method determine itself reduces Study suggests more viability.
